Die Vakzinierung gegen Tumorerkrankungen, basierend auf dem Vorhandensein von spezifischen Tumorantigenen, ist seit Jahrzehnten Gegenstand der präklinischen und klinischen Forschung. Es bestehen allerdings eine Reihe von Herausforderungen, die bis heute noch nicht überwunden sind. Hierzu zählen die Wahl des Tumorantigens, die Art der Vakzine, die Überwindung der Immunzell-Erschöpfung und der Einfluss immunsuppressiver Faktoren des Tumors selbst. Neuere Entwicklungen, welche sowohl eine Optimierung des Impfstoffes gestatten als auch neue Möglichkeiten bieten, das Tumor-Microenvironment und die Tumor-Immunevasion zu beeinflussen, geben allerdings Grund zur Hoffnung, dass Tumorvakzinen in Zukunft bei aktiven Tumorerkrankungen eine Rolle spielen könnten. Die folgende Übersicht soll einen kleinen Abriss geben, welche technischen Fragen sich an die Tumorvakzinierung stellen und welche Antworten darauf in Zukunft gegeben werden.

Vaccinations against malignant dis­eases, based on the principle of attacking specific tumor antigens, have been investigated in preclinical and clinical contexts for decades. There are, however, a variety of challenges, which until recently it has not been possible to overcome, including choice of the tumor antigen, type of vaccine as well as overcoming immune-cell exhaustion and the influence of immunosuppressive factors released by the tumor itself. Newer developments now permit the optimization of vaccines and offer possibilities to influence the microenvironment of and the immune evasion by the tumor, thereby nourishing optimism for a future role of tumor vaccines in the treatment of ac­tive malignant diseases. This article is trying to give a short overview concerning the technical questions connected with tumor vaccinations as well as answers which can possibly be given in the future.
